Level of course: basic Course objectives: The purpose is to familiarize students with the general principles for growth,evaluation and classification of microorganisms. During laboratory practice students will learn basic microbiological laboratory techniques. Prerequisites: bacis knowledge of general biology, general chemistry and biochemistry |

Laboratory sessions: 1. The rules of practice and principles of work safety in the laboratory. Basic glass and equipment for microbiological lab. Microbiological methods of sterilization.Media and methods of microorganisms cultivation.Microbiological observations. 2. Isolation of microorganisms from different sourses (water, soil). Methods of inoculation on solid medium. 3. Microscopic observation of different forms of microorgannisms. Isolation of pure culture. 4. Morphology of bacteria. Staining methods. 5. Morphology of fungi cells. Observation of yeast and mold cells 6. Detection of redox and proteolytic activity of bacteria. Bacterial activity and lactic acid fermentation 7. Rapid biochemical and serological methods for pathogenic bacteria assessment. Demonstration of Koch's postulates. 8. Detection of bacteria in selected food products. 9. Effects of temperature and selected chemicals on the growth of microorganisms. 10. Interactios between microorganisms. Types of indirect impacts. 11. Determination of microbial sensivity to antibiotics using disks diffusion methods and agar well diffusion plate methods. 12. Sewage and their purification. Microbiological analysis of the water using a membrane filtration methods. Microscopic analysis of activated sludge from wastewater. 13. Methods for quantification of bacteria and fungi. The microbiological analysis of air by Koch sedimantetion method. 14. Final exam 15. Corrective test Assesment methods: Final exam consisting of combination of multi-choice and short written answer |

Knowledge: Lo 1 - student be able to understand basic microbiological phenomena and process Lo 2 - student has knowledge of basic conceptual categories and microbiological terminology and is familiar with the development of microbiology and the research methods used in it Lo 3 - student has knowledge of the basic techniques and research tools used in microbiology Lo 4 - student knows the basic principles of health and safety in microbiological laboratory Skills: Lo 5 - student uses the basic techniques and research tools used in the mucrobiological laboratory Lo 6 - student performs simple experiments with microorganisms in the laboratory under the supervision of a scientific supervisor Competencies: Lo 7 - student is able to interact and work in a group, taking on different roles Subject effects for lecture (EK 1-2), subject effects for exercises (EK 3-7) Student activity student workload in hours participation in lecture 30 participation in laboratory 45 preparation for laboratories 5 time to write the report 15 prepare for the test 15 prepare for the exam 20 TOTAL HOURS 130/30(25) hrs. ≈4 Number ECTS: 4 |

Knoledge (Learning outcomes 3-4) Grade 2 (fail): student has no knowledge of the basic techniques and research tools used in the microbiology laboratory, knows to a very limited extent the basic principles of occupational health and safety in a microbiological laboratory Grade 3 (sufficient): student has limited knowledge of basic techniques and research tools used in the microbiological laboratory, knows the basic principles of occupational health and safety in the microbiological laboratory to a limited extent Grade 4 (good): student has good knowledge of basic techniques and research tools used in the microbiological laboratory, knows the basic principles of occupational health and safety in the microbiological laboratory Grade 5 (excellent): the student has a very good knowledge of the basic techniques and research tools used in the microbiological laboratory, knows very well the basic principles of occupational health and safety in a microbiological laboratory Skills ( Learning outcomes 5-6) Grade 2 (fail): the student uses the basic techniques and research tools used in the microbiological laboratory to a much limited extent, performs simple experiments with microorganisms under the supervision of a scientific supervisor with great difficulty Grade 3 (sufficient): the student uses the basic techniques and research tools used in the microbiological laboratory to a limited extent, with difficulty performs simple experiments with microorganisms in the laboratory under the supervision of a scientific supervisor Grade 4 (good): student is able to properly use the basic techniques and research tools used in the microbiological laboratory, knows how to perform simple experiments with microorganisms in the laboratory under the guidance of a scientific supervisor Grade 5 (excellent): student is able to very well use the basic techniques and research tools used in the microbiological laboratory, he can very well perform simple experiments with microorganisms in the laboratory under the guidance of a scientific supervisor Competencies (Learning outcomes 7) Grade 2 (fail): student is unable to interact and work in a group, taking on different roles Grade 3 (sufficient): student is able to interact to a limited extent and work in a group, taking on different roles in it Grade 4 (good): student is able to interact well and work in a group, taking on different roles Grade 5 (excellent): student is able to interact very well and work in a group, taking on different roles in it The final grade of the laboratory include: evaluation of activities and evaluation of the written test. At the final evaluation will be taken into account: 1) prepare a report after every experiment (1 - 4 points for each report). When commissioning a report late loss of some points 2) evaluation of the test (written test) 0-30 points.; 16-18 points - 3.0; 19-21pkt. - 3.5; 22-24 points. - 4.0; 25-27 points. - 4.5; 28-30 points. - 5.0 Acceptable two excused absences. |
